Do you have to run the ground wire from the DC to DC charger (12-12 30) to the negative of the battery or can I just land it to chassis ground in the back of the van and just run the positive wire to the battery with a fuse?

Chassis ground is perfect.
I do the same in my Sprinter and Berlingo.
The chassis is the best MINUS conductor yoz can get in a vehicle.
However, it us subject to corrosion if you remove the paint, hence before you screw on your MINUS wire use enough Contact Greese (e.G. ENSTO) to seal the whole hole and area.
